RESUMEN:Salmonella enterica es un patógeno zoonótico distribuido mundialmente, con importantes efectos negativos en salud animal y pública. En Uruguay, la actividad agropecuaria es central en la economía. La salmonelosis bovina puede producir importantes pérdidas para la ganadería, y los bovinos actuar como fuente de infección humana y reservorio de cepas con resistencia antibiótica. En este trabajo se evaluó la asociación entre la infección por Salmonella enterica y el síndrome de diarrea neonatal bovina, los factores asociados a la infección y la mortalidad de terneros en tambos de Uruguay. Además, se determinó el serotipo y perfil de sensibilidad antibiótica de los aislamientos de Salmonella enterica obtenidos. Se evaluaron clínicamente y se tomaron muestras de heces de 544 terneros de 1 a 30 días de edad, incluyendo 262 terneros con y 282 terneros sin diarrea, en un total de 30 tambos (estudio I). A su vez, se analizaron por bacteriología e histopatología muestras de tejidos y/o fluidos corporales de terneros necropsiados procesados en la Plataforma de Investigación en Salud Animal de INIA La Estanzuela, para determinar la causa de muerte (estudio II). En el estudio I se aisló S. enterica en 22 (4,04%) muestras de heces de terneros que pertenecían a 9/30 establecimientos (30%). A su vez, en el estudio II, se aisló S. enterica en 19 casos de 12 brotes de mortalidad de terneros y se vinculó el aislamiento con los hallazgos patológicos macro y/o microscópicos, que fueron típicos de salmonelosis. Salmonella Typhimurium fue el serotipo más frecuentemente aislado de heces (19 aislamientos, 86,4%), seguido de S. Anatum (3 aislamientos, 13,6%), en el estudio I. En 10 de los 12 brotes de mortalidad estudiados (estudio II) se detectó el serotipo S. Typhimurium (83,3%), mientras que S. Dublin se detectó en los restantes 2 (16,7%). En el estudio I hubo asociación estadísticamente significativa entre la infección y varios parámetros clínicos, entre los que se destacaron: actitud anormal, reducción del reflejo de succión y capacidad de incorporación de los terneros, presencia de fibrina en las heces y fiebre. Del panel de 15 antibióticos evaluados, los aislamientos de Salmonella enterica obtenidos de ambos estudios presentaron mayor frecuencia de resistencia frente a estreptomicina (38/41), tetraciclinas (36/41) y ciprofloxacina (15/41), y todos fueron sensibles a fosfomicina. La presencia de aislamientos multirresistentes fue frecuente (34/41 aislamientos evaluados, 82,9%), encontrándose aislamientos con sensibilidad disminuida frente hasta seis grupos de antibióticos. Se concluye que S. enterica está asociada a enfermedad neonatal y mortalidad de terneros en establecimientos lecheros de Uruguay, que S. Typhimurium es el serotipo encontrado con mayor frecuencia, y que hay cepas de Salmonella enterica de origen bovino que presentan resistencia/multirresistencia a antibióticos. El diagnóstico clínico, patológico y bacteriológico de salmonelosis representa la base para comprender la epidemiología de esta enfermedad, implementar medidas preventivas y terapéuticas, como así también estudiar en profundidad las características microbiológicas de las cepas circulantes en el país.
SUMMARY: Salmonella enterica is a zoonotic pathogen of worldwide distribution, with important effects on animal and public health. Livestock farming is central to the economy of Uruguay. Bovine salmonellosis can cause important economic losses to the livestock industry, and cattle can serve as source of human infections, and reservoirs of Salmonella strains with antibiotic resistance. In this work, we assessed the association between Salmonella enterica infection and neonatal calf diarrhea syndrome, factors associated with the infection, and calves´ mortality in dairy farms in Uruguay. Additionally, the Salmonella enterica serotypes infecting cattle and their antibiotic sensitivity profile were determined. A total of 544 neonate dairy calves from 1 to 30 days of age from 30 dairy farms, including 262 calves with diarrhea and 282 calves without diarrhea, were clinically evaluated, and individual fecal samples were collected (study I). Furthermore, samples of tissues and/or body fluids obtained from necropsied calves were processed at INIA La Estanzuela Animal Health Platform for histopathology and/or bacteriology to investigate the cause of death (study II). In study I, S. enterica was isolated from 22 (4.04%) fecal samples from calves from 9 (30%) farms. In study II, S. enterica was isolated in 19 cases in 12 spontaneous outbreaks of calf mortality, and the isolation was linked to the gross and/or microscopic pathological findings in the necropsied calves, that were typical of salmonellosis. Salmonella Typhimurium was the most frequent serotype isolated from feces (19 isolates, 86.4%), followed by S. Anatum (3 isolates, 13.6%) in study I. In 10 of the 12 mortality outbreaks studied (study II), S. Typhimurium was detected (83.3%), while S. Dublin was found in the remaining 2 (16.7%). The factors associated with Salmonella infection were determined in study I. There was an association between the infection and several clinical parameters, including abnormal attitude of the calves, abnormal suction reflex, reduced calves´ ability to stand up, presence of fibrin in the feces, and fever. Antibiotic sensitivity testing using a panel of 15 antibiotics, revealed that the Salmonella enterica isolates showed higher frequency of resistance to streptomycin (38/41), tetracyclines (36/41) and ciprofloxacin (15/41), while all isolates were sensitive to fosfomycin. Thirty-four of the 41 isolates (82.9%) assayed were resistant to multiple antibiotics (multirresistant strains), with some isolates showing decreased sensitivity to up to six groups of antibiotics. We conclude that Salmonella enterica is associated with neonatal calf disease and calf mortality in dairy farms in Uruguay, that S. Typhimurium is the most frequently isolated serotype, and that there are strains of Salmonella enterica of bovine origin that show resistance/multirresistance to antibiotics. The clinical, pathological and bacteriological diagnosis of salmonellosis represents the basis for understanding the epidemiology of this disease, implementing preventive and therapeutic measures, and further studying the microbiological characteristics of Salmonella strains in Uruguay. Typhimurium es el serotipo encontrado con mayor frecuencia, y que hay cepas de Salmonella enterica de origen bovino que presentan resistencia/multirresistencia a antibióticos. El diagnóstico clínico, patológico y bacteriológico de salmonelosis representa la base para comprender la epidemiología de esta enfermedad, implementar medidas preventivas y terapéuticas, como así también estudiar en profundidad las características microbiológicas de las cepas circulantes en el país. SUMMARY: Salmonella enterica is a zoonotic pathogen of worldwide distribution, with important effects on animal and public health. Livestock farming is central to the economy of Uruguay. Bovine salmonellosis can cause important economic losses to the livestock industry, and cattle can serve as source of human infections, and reservoirs of Salmonella strains with antibiotic resistance. In this work, we assessed the association between Salmonella enterica infection and neonatal calf diarrhea syndrome, factors associated with the infection, and calves´ mortality in dairy farms in Uruguay. Additionally, the Salmonella enterica serotypes infecting cattle and their antibiotic sensitivity profile were determined. A total of 544 neonate dairy calves from 1 to 30 days of age from 30 dairy farms, including 262 calves with diarrhea and 282 calves without diarrhea, were clinically evaluated, and individual fecal samples were collected (study I). Furthermore, samples of tissues and/or body fluids obtained from necropsied calves were processed at INIA La Estanzuela Animal Health Platform for histopathology and/or bacteriology to investigate the cause of death (study II). In study I, S. enterica was isolated from 22 (4.04%) fecal samples from calves from 9 (30%) farms. In study II, S. enterica was isolated in 19 cases in 12 spontaneous outbreaks of calf mortality, and the isolation was linked to the gross and/or microscopic pathological findings in the necropsied calves, that were typical of salmonellosis. Salmonella Typhimurium was the most frequent serotype isolated from feces (19 isolates, 86.4%), followed by S. Anatum (3 isolates, 13.6%) in study I. In 10 of the 12 mortality outbreaks studied (study II), S. Typhimurium was detected (83.3%), while S. Dublin was found in the remaining 2 (16.7%). The factors associated with Salmonella infection were determined in study I. There was an association between the infection and several clinical parameters, including abnormal attitude of the calves, abnormal suction reflex, reduced calves´ ability to stand up, presence of fibrin in the feces, and fever. Antibiotic sensitivity testing using a panel of 15 antibiotics, revealed that the Salmonella enterica isolates showed higher frequency of resistance to streptomycin (38/41), tetracyclines (36/41) and ciprofloxacin (15/41), while all isolates were sensitive to fosfomycin. Thirty-four of the 41 isolates (82.9%) assayed were resistant to multiple antibiotics (multirresistant strains), with some isolates showing decreased sensitivity to up to six groups of antibiotics. We conclude that Salmonella enterica is associated with neonatal calf disease and calf mortality in dairy farms in Uruguay, that S. Typhimurium is the most frequently isolated serotype, and that there are strains of Salmonella enterica of bovine origin that show resistance/multirresistance to antibiotics.
